My two favorite tips..

1) To kill a layer without being asked if you are sure etc.:
Shift + Alt + while clicking the trash can.

2) To make a flatened layer of all visible layers, while still retaining all layers intact. :
Shift + Alt while pressing Merge layers.

It adds a flatened version to your layers pallet.

Here's a variation on tip number 2....

Create a new layer
Cntrl + Shift + n

then

Cntrl + Shift + Alt + e
creates a merged version of all layers on the new layer
