Are you making the images for the posters yourself or are you using images you already have such as Photographs?

If you are making the graphics yourself then I suggest you use Adobe Illustrator and make them in vector format so that you can print them at any size you want without losing quality.

If you are usign Photographs then I hope they are very high resolution, or else they are going to get very pixelated when you blow them up this big.

If you are making these large files at 300dpi in Photoshop and not Illustrator then I hope you have a lot of memory in your computer, you will need it
