Make the opacity layer back to 100% and the Fill layer also.

Select all three layers (shift+ctrl+click on each thumbnail)

Create a new layer as bottom layer (select fill 50% layer and press and hold ctrl+click new layer icon)

Fill the new masked bottom layer with white while the three layers are selected (marching ants)

Make the fill layer back to 50 % and the Opacity layer also.

Deselect all and save as PNG.

It is important to start with all 3 layers at fill and opacity 100% because otherwise it will affect the white bottom layer.
