Thanks for response, i am under impression that saving can affect quality of image since its not lossless (with rectangular boxes its not visible, but if it was more complicated image, it can potentially make it more pixelated or blurred).

I managed to eliminate step with saving a and opening PNG by merging down "opacity 50%" and "fill 50%" layers on the white background created right under them and new layer was not transparent anymore (since it has solid white background under transparent object).
