    If you are creating your art in Illustrator and not adding Raster Images your file will be Vector Art and can be scaled to any size without quality lost. Seeing that is to be Screen Printed I would just create them at the largest size.

    Using the Pen Tool would be the best way to get clean lines.

    If you could Post the image it will give us a better idea of what your trying to do. Is this going to be Digitally printed or Offset printed? If it is Offset you are going to have Trapping issues with the 877.

    200 to 300% is the Zoom shown at the top off your image.(How much it is blown up on your screen) Make your Path into the light part by a couple of Pixels.
